AMPK helps coordinate metabolic processes in response to a cell’s energy status. Researchers study berberine partly because experimental findings connect it with AMPK-related signaling and glucose and lipid metabolism.
PRECLINICAL / MECHANISTIC RESEARCHWhat about mitochondria and ATP?
Research in aging rats examined AMPK/SIRT1/PGC-1α signaling and mitochondrial outcomes after berberine treatment. Such experiments help identify mechanisms worth studying. They do not establish that Burn increases ATP, creates new mitochondria, or causes fat loss in people.

A review of randomized placebo-controlled trials found reductions in fasting glucose, triglycerides and waist circumference in the studied populations. LDL cholesterol, total cholesterol and BMI also improved; HDL and blood pressure did not show significant changes. The authors called for higher-quality trials.

A meta-analysis of 23 articles reported an average weight difference of −0.88 kg (about 1.9 lb) versus control groups, alongside changes in BMI and waist circumference. Waist-to-hip ratio did not significantly change. Published online in November 2025, in the January 2026 journal issue. Results varied across study protocols.

This review involved people with type 2 diabetes. It reported improvements in several glucose and lipid measures, with different findings for berberine alone and berberine added to medication. Common doses were 900–1,500 mg daily for one to three months. Those populations and doses differ from Burn’s intended daily use; this is not guidance to treat diabetes or change medication.

In 337 adults with obesity and fatty liver disease without diabetes, six months of berberine did not significantly reduce visceral or liver fat compared with placebo. This helps explain why a promising mechanism is not a guarantee of a particular clinical outcome.
